Vitamin B12 Shots:

The main benefit of Vitamin B12 shots is that theyre rapidly absorbed from intramuscular sites of injection; the plasma level of the compound reaches its peak within one hour after intramuscular injection.

As an example, if you took a Vitamin B12 pill that contained 500 micrograms of B12 (which is more than most), then your body would (on average) only absorb about 1.8 micrograms of that B12 which is 0.3% of what you thought you were taking!  This is why so many people have low-levels of Vitamin B12.

When the B12 vitamin enters your body via the stomach (through food ), acids in the stomach separate the B12 from its protein source.  It then must combine with intrinsic factor cells in the stomach (this is a protein created by the body except in people with Pernicious anemia).  This B12/intrinsic factor complex travels through your intestines where it is absorbed.  The absorbed amount is then transported via blood plasma and stored in the liver.  This process is inefficient even in healthy humans which explains why so many adults have some level of vitamin B12 deficiency.
